- 非典型抗精神病藥，所以作用機轉不是D2 receptor competitive antagonist，而是D4 receptor competitive antagonist和block of 5-HT2A autoreceptors，因此不像典型抗精神病藥有椎體外症候群。
- Clozapine has high affinity for D1, D2, D4, 5-HT2,muscarinic, and a-adrenergic receptors,but it is also a dopamine D2 receptor antagonist.
治療學[]
- negative symptom of 思覺失調症(舊名精神分裂症)，或是對typical one沒有反應的positive symptom.
- Risperidone should be included among the first-line antipsychotic drugs, whereas Clozapine is reserved for treatment of individuals who are unresponsive to other neurloleptics, because its use is associated with blood dyscrasias
毒性[]
血液毒性:惡血質(blood dyscrasias):Clozapine can produce bone marrow suppression and cardiovascular side effects. The risk of severe agranulocytosis necessitates frequent monitoring.
